A MALE MOUSE MODEL OF REPRODUCTIVE IMPAIRMENT INDUCED BY LONG-TERM HIGH-DOSE PARACETAMOL

Abstract

Background: Prolonged use of paracetamol can cause endocrine disruption and affect reproductive function in men. Objectives: To evaluate semen parameters in mice treated with longterm high-dose paracetamol intervention. Materials and methods: Experimental studies were conducted on 48 male Swiss albino mice, 5-6 weeks old, 22±2 g, randomly divided into 4 groups: physiological control (group 1), positive control (group 2) receiving 750 mg/kg/day of sodium valproate orally, and intervention groups (groups 3 and 4) receiving 300 or 400 mg/kg/day of paracetamol intravenously. Semen analysis was performed after 10 or 15 weeks of intervention. Results: After 15 weeks, sodium valproate (positive control) induced a marked deterioration in sperm quantity and quality compared with the physiological control group following daily administration during the first 5 weeks, with significantly reduced sperm concentration (1.61±1.32 million/mL), decreased motility (4.58±2.13%), and increased abnormal morphology (40.34±3.49%) (p<0.001). Paracetamol at a dose of 300 mg/kg did not produce significant changes in semen parameters compared with the physiological control after 10 weeks, but there were significant abnormalities, including reduced sperm concentration (51.64±1.65 million/mL) and motility (82.06±2.02%), as well as increased immotility (17.94±2.02%) and abnormal morphology (19.01±2.54%) after 15 weeks (p<0.001). Paracetamol at 400 mg/kg resulted in reductions in sperm concentration and motility as early as 10 weeks, with more pronounced impairment characterized by sperm concentration (38.41±3.03 million/mL), motility (39.41±5.28%), immotility (60.59±5.28%), and abnormal morphology (30.31±4.60%) after 15 weeks (p<0.001). Conclusions: Long-term use of paracetamol at a dose of 400 mg/kg resulted in impaired fertility in male white mice after 15 weeks.
