Abstract

Background: Products of biological origin effective response in the treatment of burns and skin lesions loss is a matter of urgency. we proceed the study:“Create keratinocytes plate from cord blood stem cells on the substrate amnion”. Objectives: Create keratinocytes plate with multiple layers on the substrate amnion. Materials and method: The study was designed experimental description. Subjects of study were cord blood and amniotic membranes from the placenta of a full-term newborn. We collected mesenchymal stem cells from umbilical cord blood, identify and cultivate cell proliferation. Receiving and processing of the amniotic membrane into a carrier cell. Mesenchymal stem cell transplantation from umbilical cord blood to amnion. When the cell density reaches 60-80% of the area, we performed differentiation of mesenchymal stem cells into skeratinocytes. Observe under the reverse microscope to record the change in cell shape. Cell shape change reached 80%. We conducted the cascading process by airlifting method. The airliffting process was done in 7 days. Products evaluated: morphology (HE), expression of P63, CK 5/6, and TEM. Result: Successfully obtained mesenchymal stem cells from umbilical cord blood. Receive and treat the membranes successfully into cell-bearing media. Successfully create keratinocytes plate with 3-5 layers of cells. Conclusion: Successful generation of keratinocytes plate with multiple layers on the substrate amnion.
