THE HISTOPATHOLOGICAL CHARACTERISTICS AND EXPRESSION STATUS OF MLH1, MSH2, MSH6, PMS2 PROTEINS IN COLORECTAL ADENOCARCINOMA AT CAN THO ONCOLOGY HOSPITAL IN 2019-2020

Abstract
Background: In colorectal adenocarcinoma, it is critical to determine the expressions of mismatch repair proteins that are strongly associated with the prognosis and the response prediction to 5-FU chemotherapy. Objectives: 1) to determine the rates of the histopathological characteristic of colorectal adenocarcinoma at Can Tho Oncology Hospital from 2019 to 2020; 2) to determine the percentage of loss expression of MLH1, MSH2, MSH6, PMS2 proteins, as well as their involvement with the histopathological characteristic of colorectal adenocarcinoma at Can Tho Oncology Hospital from 2019 to 2020. Materials and Methods: A cross-sectional descriptive study was conducted on 131 patients diagnosed as colorectal adenocarcinoma via HE staining, included from January 2019 to July 2020. The Immunohistochemistry technique was carried out of the 5µm slices of patient's tumor tissue to determine the MLH1, MSH2, MSH6, PMS2 proteins expressions at the Anapathology department – Can Tho Oncology Hospital. Results: On 131 colorectal adenocarcinoma patients, the adenocarcinoma type accounted almost for 90.1%, and mucous or ring cell and micropapillary adenocarcinoma type accounted for 9.9 %. The low-grade carcinoma and high-grade carcinoma were respectively 92.4% and 7.6%. The rate of dMMR was 36.6% and the rates of loss expression of each protein types by immunohistochemistry stained technique were 26% for MLH1, 16.8% for MSH2, 17.6% for MSH6 and 22.1% for PMS2, respectively. There was not a significant correlation between the loss of proteins expression with pathohistological type, and pathohistological grade. Conclusion: The adenocarcinoma was the major type of colorectal adenocarcinoma (90.1%). Almost tumors have had histological low-grade (92.4%). The rate of loss of MLH1 protein was highest (26%), whereas the rate of loss of MSH2 protein was lowest (16.8%). The total rate of dMMR was 36.6%. There was no significant correlation between the loss of MSH2, MSH6, PMS2 proteins expression with histopathological type and histopathological grade.
